An Ultrastructural Study of the Hepatoma Induced by 3¢¥-methyl-4-dimethylaminoazobenzene

Abstract
A total of 56 sprague Dawley male rats used for the experiment, and divided into four groups according to the sacrifice interval. Administration of 3¢¥-Me-DAB were carried out daily by nasogastric tube insertion. Animals were sacrificed serially at different interval after administration of 3¢¥-Me-DAB for 10 weeks. At the time of sacrifice, the induced tumors and a portion of caudate lobe was prepared for the histological and electron microscopic observation by routine procedures. Patholgocally most of the induced tumors were cholangiocarcinoma, and a small numbers of hepatocellular carcinoma and mixed tumor were alse induced. Ultrastructurally the induced cholangiocarcinoma showed small numbers of mitochondria and endoplasmic reticulum in the cytoplasm and different stages of microvilli formation in the cell membrane. In hepatocellular carcinoma, small sized mitochondria and proliferation of rough endoplasmic reticulum was frequentaly observed. In mixed tumor, both of the two types of cells were mixed in the same lesion. Based on these results, most of the hepatomas induced by 3¢¥-Me-DAB are cholangiocarcinoma and ultrastructurally oval cells are believed to the progenitor cell of this tumor.
